Nitrous oxide speeds the reduction of distressing intrusive memories in an experimental model of psychological trauma

Inhaling nitrous oxide shortly after a traumatic event speeds the decline of intrusive memories, reducing them by the next day compared to four days later with medical air. The gas appears to disrupt memory consolidation, but it increases intrusive memories in people who were highly dissociated at the time of the trauma. Sleep quality did not differ between groups. The findings suggest nitrous oxide may help prevent PTSD symptoms in some individuals, but caution is warranted for those with dissociation.

MDMA (ecstasy) and compassionate imagery both increase self-compassion and reduce self-criticism in recreational users. In a non-blind experiment, participants who consumed ecstasy showed similar pro-social effects to those produced by a contemplative compassion exercise, particularly in those with higher attachment-related avoidance. The findings suggest MDMA may enhance psychotherapy by fostering compassionate attitudes toward oneself. However, because the study was not blinded and drug purity was unknown, controlled trials with pharmaceutical-grade MDMA are needed to confirm these effects.

In a naturalistic setting, recreational users who consumed chemically verified MDMA performed a self-focused compassionate imagery exercise on two separate occasions: once after taking MDMA and once without. The exercise and MDMA each independently increased emotional empathy toward critical facial expressions and self-compassion, and their effects on self-compassion appeared to add together. The findings suggest that combining MDMA with compassionate imagery may enhance self-directed affiliative feelings, though controlled studies are needed to confirm the combined utility for therapeutic contexts.

A new 20-item Psychedelic Preparedness Scale (PPS) was developed and validated to measure how well participants are prepared for psychedelic experiences. Using an iterative Delphi-focus group method and input from clinicians, researchers, and psychedelic users, the scale identifies four factors: Knowledge-Expectations, Intention-Preparation, Psychophysical-Readiness, and Support-Planning. Pharmacological modelling of dissociation and psychosis: an evaluation of the Clinician Administered Dissociative States Scale and Psychotomimetic States Inventory during nitrous oxide (‘laughing gas’)-induced anomalous states

Nitrous oxide (N2O) produces dissociative and psychosis-like effects comparable to those of ketamine in healthy volunteers, making it a practical alternative for modeling these experiences outside clinical settings. Analysis of data from three previous studies found that the Clinician Administered Dissociative States Scale (CADSS) and Psychotomimetic States Inventory (PSI) capture largely non-overlapping subjective experiences during N2O inhalation. A three-factor model of dissociation was confirmed, though a two-factor model may be more parsimonious. Psychosis-like symptoms were represented by two negative and two positive symptom factors. The findings suggest both measures should be used together to comprehensively assess anomalous states from dissociative NMDAR antagonists.

Inhaling a high dose of vaporized synthetic 5-MeO-DMT radically reorganizes low-frequency brain oscillations, making them heterogeneous, viscous, and nonrecurring, and halting their typical forward and backward travel across the cortex. This reorganization also causes broadband neural activity to become more stable and low-dimensional, with increased energy barriers for rapid global shifts. These findings, based on EEG data from 29 healthy individuals, provide a detailed account of how the drug sculpts human brain dynamics and reveal atypical cortical slow-wave behaviors relevant to neuroscientific models of serotonergic psychedelics.

Perceived changes in mental health and social engagement attributed to a single psychedelic experience in autistic adults: results from an online survey.

The majority of autistic participants who completed an online survey reported that their most impactful psychedelic experience reduced psychological distress (82%) and social anxiety (78%) and increased social engagement (70%). A substantial minority (20%) reported undesirable effects such as increased anxiety, with some describing the experience as among the most negatively impactful of their lives. The only substantial predictor of reduced distress was increased psychological flexibility. The findings come from a non-experimental design with biased sampling, so caution is warranted.

A high dose of the psychedelic drug 5-MeO-DMT radically reorganizes low-frequency brain activity in 29 healthy individuals. Inhaling 12 mg of vaporized synthetic 5-MeO-DMT caused neural activity flows to become incoherent, heterogeneous, viscous, fleeting, and nonrecurring, ceasing typical traveling waves across the cortex. This reorganization led to slower, more stable, low-dimensional broadband activity with increased energy barriers to rapid global shifts. The findings provide the first detailed empirical account of how 5-MeO-DMT alters human brain dynamics, revealing novel cortical slow wave behaviors.

The Induction of Dissociative States: A Meta-Analysis.

Dissociative states—disruptions in awareness and perception—occur across many psychiatric conditions and can be modeled in the lab. A meta-analysis of 123 studies (6,692 individuals) measured state dissociation using a standardized scale. At baseline, the largest effects were in dissociative and complex subtypes of posttraumatic stress disorder. In controlled experiments, mirror gazing and several drugs, particularly ketamine and cannabis, induced dissociation as high as or higher than that seen in PTSD. Results were highly variable across studies but not explained by methodological differences. These findings validate experimental methods for inducing dissociation and inform monitoring of adverse events in drug-based interventions.

Personalized high-precision neurofeedback (NF) can help novice meditators better disengage from mental activity during meditation, improving emotional well-being and mindful awareness. In a single-blind, controlled study, 40 novices received two days of meditation training with feedback from either their own or a matched participant's posterior cingulate cortex (PCC) activity, measured using 7 Tesla fMRI. The experimental group showed stronger functional decoupling of PCC from dorsolateral prefrontal cortex, indicating improved control over disengagement. This led to greater improvements in emotional well-being and mindful awareness during a week of real-world self-guided meditation, supporting the utility of NF-guided meditation training.

This review brings together major UK psychedelic research teams to formalise these unique considerations, addressing sociocultural, political, legal, pharmacological, safety, study design, and experiential facets. It identifies continuing areas of debate and provides a practical, experience-based guide with recommendations for policymakers and future researchers intending to set up a psychedelic study or clinical trial.

Practitioner perspectives on meditation-based preparation for psychedelic experiences

Experienced meditators who also use psychedelics expect meditation to enhance acceptance and present-moment focus during psychedelic sessions, while increasing feelings of oceanic boundlessness and reducing anxious ego-dissolution. Among those with limited meditation experience (1-3 years) and few psychedelic sessions (1-20), adhering to a specific meditation tradition was linked to higher perceived benefits of meditation for psychedelic preparation. However, for highly experienced meditators (10+ years) with limited psychedelic use, non-adherents reported higher perceived benefits. Loving-Kindness Meditation was rated the most beneficial preparatory practice. Participants recommended about three weeks of daily 30-minute sessions, favoring online and asynchronous delivery.

Development and validation of a short-form (6-item) version of the clinician-administered dissociative states scale (CADSS-SF).

A 6-item short form of the Clinician Administered Dissociative States Scale (CADSS-SF) was developed using data from three studies of nitrous oxide in 229 healthy volunteers, then validated in 80 separate participants. The single-factor scale, composed of derealization and depersonalization items, showed excellent model fit and internal consistency (omega = 0.87), correlated strongly with the full 19-item CADSS (r ≥ 0.88), and moderately with a measure of psychotomimesis (r = 0.63). The CADSS-SF enables rapid, repeated assessment of dissociation during drug intoxication without disrupting the experience, but primarily captures derealization and depersonalization and requires further validation beyond drug-induced dissociation in healthy populations.

Protocol for a qualitative mechanistic study of MDMA with a sample of psychoanalytic psychotherapists: A phenomenological investigation

MDMA-assisted psychotherapy shows promise for treating PTSD, but the psychological mechanisms behind its effects are not well understood. This qualitative study will have approximately 25 experienced psychodynamic psychotherapists receive two doses of MDMA (80-120 mg, oral) in an open-label design. Participants will complete interviews before, during, and after MDMA sessions, plus daily journals. Researchers will use phenomenological and thematic analysis, along with grounded theory, to develop a model of MDMA's psychological mechanisms of action. The study treats therapists as expert observers of their own psychological processes, aiming to generate insights that can inform future treatment models and offer a framework for qualitative mechanistic research on psychedelics.

A 21-day mobile-accessible programme called the Digital Intervention for Psychedelic Preparation (DIPP) is being tested for feasibility and preliminary efficacy in a randomised controlled trial. The study will recruit 40 non-treatment-seeking adults without a clinical diagnosis, randomly assigning them to either a guided meditation with music condition or a music-only condition. After the digital intervention, all participants will attend an in-person supervised psilocybin session with a standardised 25 mg dose. Primary outcomes include recruitment efficiency, retention, and adherence; secondary outcomes assess preparedness, quality of the psychedelic experience, and wellbeing, with follow-ups up to 9 months. The trial is registered as NCT06815653.

Investigating the Role of Self-Compassion in MDMA-Assisted Therapy for Social Anxiety Disorder

Self-compassion is thought to be important for mental health, especially for people with social anxiety disorder. MDMA-assisted therapy may work by increasing self-compassion. This study will examine whether trait self-compassion rises during treatment and whether self-compassion felt during MDMA dosing sessions leads to lasting changes. It will also test if changes in self-compassion relate to improvements in social anxiety, depression, shame, functioning, and belonging. Five specific aims are outlined to explore these relationships across two dosing sessions.

This registered clinical study investigates whether N,N-dimethyltryptamine (DMT), alone or combined with reactivating alcohol-related memories, can produce lasting changes in the brain, cognition, and drinking behavior in people with mild alcohol use disorder who are hazardous drinkers but not seeking treatment. Up to 120 participants will be assigned to one of four groups: alcohol memory retrieval plus DMT, alcohol memory retrieval plus placebo, control memory retrieval plus DMT, or control memory retrieval plus placebo. Drinking levels will be measured over three lab sessions and a nine-month follow-up using timeline follow-back and blood tests.

Electroceutical enhancement of self-compassion training using transcutaneous vagus nerve stimulation: results from a preregistered fully factorial randomized controlled trial

Transcutaneous vagus nerve stimulation (tVNS) combined with a brief self-compassion mental imagery training (SC-MIT) produced a larger acute increase in state self-compassion and mindfulness than either intervention alone in healthy adults. Over eight daily sessions, tVNS plus SC-MIT showed accumulating effects on mindfulness but not self-compassion. Self-criticism and attentional bias toward compassionate faces were influenced only by the type of imagery training, not by stimulation. Heart rate variability was unaffected by any condition. The findings suggest tVNS may enhance meditation-based psychotherapies, while oculomotor metrics appear sensitive to self-compassion training.

Strategies for resolving challenging psychedelic experiences: insights from a mixed-methods study.

People who have difficult experiences while using psychedelics can benefit from specific coping strategies. A mixed-methods study of psilocybin retreat participants and a larger online survey identified three main ways people navigate challenging psychedelic experiences: accepting and reinterpreting the experience, regulating through sensory and physical interaction, and seeking social support. Acceptance and reappraisal, along with social support and disclosure, were linked to greater emotional breakthrough. Fear-related challenges were associated with less emotional breakthrough and fewer adaptive coping strategies. These findings clarify how challenging experiences can lead to positive outcomes and suggest ways to improve therapeutic protocols and safety.
