Acute effects of nitrous oxide on visual processing: a connectome study in healthy adults.

Subanesthetic nitrous oxide (N2O) alters visual experience by reconfiguring large-scale brain networks rather than changing early visual processing. In a placebo-controlled fMRI study with 13 healthy adults, participants viewed a flashing checkerboard and rated visual intensity and unpleasantness. Increased unpleasantness under N2O was linked to reduced connectivity between the right anterior insula and the anterior cingulate cortex and lateral occipital cortex. Network analyses revealed reduced modularity and a collapse of hierarchical organization, with sensorimotor connectivity redistributed toward salience and associative networks. These findings suggest that altered visual experience under N2O arises from disrupted salience integration and increased cross-network communication.

Awakening to Death: How Long-Term Sufi Meditation Shapes Prosocial Responses to Mortality Salience

Distinct Neurochemical Signature of Mindfulness and Progressive Muscle Relaxation in limbic regions: a randomized controlled MR spectroscopy study

More than meets the eye: neural correlates of consciousness in the sound-induced flash illusion

The visual awareness negativity (VAN) is a brain signal thought to reflect conscious perception. This event-related potential study tested whether the VAN appears during visual illusions, where people report seeing something that is not physically present. Using the sound-induced flash illusion—a single flash accompanied by two beeps often creates the illusion of a second flash—the researchers compared brain responses from 47 participants on trials where the illusion was perceived versus not perceived. An enhanced negativity over posterior electrodes between 250 and 300 milliseconds appeared during illusory trials. Lower pre-stimulus alpha power also accompanied illusion trials, consistent with prior work linking alpha brain waves to perceptual variability. These results suggest the VAN reflects conscious perception even for illusory experiences, supporting its role as a neural correlate of consciousness.

Cannabinoid use after stress causes male rats to generalize fear responses to neutral cues and adopt avoidant coping behaviors. The combination of THC and CBD triggered distinct brain changes in the nucleus accumbens core: astrocytes retracted from synapses and Synapsin-I density decreased when a neutral odor was presented, while exposure to a stress-conditioned odor activated MMP-2,9 (mostly MMP-2), re-associated astrocytes to synapses, increased Synapsin-I density, and caused astrocyte atrophy. These neuroadaptations occurred only in males. The findings suggest MMPs and astrocytes as potential therapeutic targets for co-occurring cannabis use disorder and PTSD.

Attitudes Toward Psychedelic-Assisted Therapies for Cancer-Related Psychosocial Symptoms: A Multi- Stakeholder Analysis

A national survey of 742 Canadian stakeholders—cancer patients, healthcare providers, and policymakers—found that patients held the most favorable attitudes toward psychedelic-assisted therapy (PAT), though they reported the least knowledge about it. AI Discovery of Mechanisms of Consciousness, Its Disorders, and Their Treatment

A generative adversarial AI framework, pitting deep neural networks against interpretable neural field models, produces biologically realistic simulations of conscious and comatose brains that recapitulate features across humans, monkeys, rats, and bats. Without explicit programming, the AI retrodicts known responses to brain stimulation and generates testable predictions about unconsciousness mechanisms. Two predictions are validated: selective disruption of the basal ganglia indirect pathway, supported by diffusion MRI in 51 DOC patients; and increased cortical inhibitory-to-inhibitory synaptic coupling, supported by RNA sequencing from resected brain tissue in six human coma patients and a rat stroke model. The model also identifies high-frequency subthalamic nucleus stimulation as a promising intervention, supported by electrophysiology data from human patients.

The endogenous opioid system in the medial prefrontal cortex mediates ketamine’s antidepressant-like actions

A single dose of ketamine's antidepressant-like effects in rats require the brain's own opioid system, specifically β-endorphin and mu-opioid receptors in the medial prefrontal cortex (mPFC). Blocking opioid receptors with naltrexone, either throughout the body or directly in the mPFC, prevented ketamine's behavioral effects and associated molecular changes. Ketamine rapidly increased β-endorphin levels and mu-opioid receptor gene expression in the mPFC, as well as the gene for β-endorphin's precursor in the hypothalamus. Neutralizing β-endorphin in the mPFC with a specific antibody before ketamine also abolished both behavioral and molecular effects, demonstrating that activation of mPFC opioid receptors is necessary for ketamine's antidepressant-like actions.
