Ketamine increases redundancy in brain dynamics, particularly in the alpha frequency band, and this effect is more pronounced during resting state and associated with dissociative experiences. In a double-blinded cross-over design with 30 male adults, racemic ketamine was compared to saline infusion. Higher-order interactions (HOI) computed from EEG data showed that ketamine boosted redundancy, especially for predictable stimuli in an auditory oddball task. These findings suggest that ketamine-induced shifts toward dissociation correlate with increased redundancy in neural signal interactions, highlighting the potential of complexity measures with portable EEG for monitoring pharmacological changes in consciousness.

About one third of people with depression do not fully respond to standard treatments, and psilocybin may offer a rapid-acting alternative. This registered trial will randomize 36 adults with major depressive or persistent depressive disorder to receive either 25 mg psilocybin or an active placebo (100 mg niacin), then cross over three weeks later so that all participants receive psilocybin. Using serial neuroimaging, the study will test whether psilocybin acutely alters cerebral blood flow and functional brain activity in mood-related networks compared to placebo, and whether those changes persist subacutely. Clinical scales and serum biomarkers will also be collected to explore relationships with treatment response.

A single session of mindfulness meditation, whether 10 or 20 minutes, increases state mindfulness more than listening to a National Geographic article. Ten minutes of meditation was sufficient to produce this effect, and extending the session to 20 minutes did not yield additional gains. For people with lower trait mindfulness, meditation boosted state mindfulness more than the control. Among those with high trait mindfulness, a 20-minute session led to greater reductions in state anxiety than a 10-minute session. The minimal dose-response relationships suggest that 10 and 20 minutes of meditation improve state mindfulness to a similar degree.

Machine learning applied to 11,816 publicly available drug testimonials reveals distinct subjective experiences linked to specific substances. Using BERT, a transformer model, the study predicted 28 dimensions of sentiment across narratives, validated by psychiatrist annotations. Canonical correlation analysis connected 52 drugs' receptor affinities with word usage, uncovering 11 latent receptor-experience factors mapped to a 3D cortical atlas. Results distinguished lucid from mundane phenomena: MDMA was linked to "Love," DMT and 5-MeO-DMT to "Mystical Experiences," and other tryptamines to "Surprise," "Curiosity," and "Realization." These models could potentially guide real-time biofeedback in therapeutic sessions.

In a rat model of post-traumatic stress disorder (PTSD), N,N-dimethyltryptamine (DMT) and the combination of DMT with the monoamine oxidase inhibitor harmaline (pharmahuasca) reduced reactive oxygen species (ROS) production in the prefrontal cortex and hippocampus, while harmaline alone had mixed effects. RNA sequencing of the prefrontal cortex showed that DMT and pharmahuasca altered expression of genes related to ROS production, inflammation, neurotransmission, and neuroplasticity. DMT, but not harmaline, showed both affinity and efficacy at the human 5HT2A receptor. The findings suggest DMT and pharmahuasca may help treat PTSD by reducing oxidative stress and inflammation and promoting neuroplasticity.

LSD increases the force and rate of heart muscle contraction by activating H2-histamine receptors in humans, and also acts as a partial agonist at 5-HT4 serotonin receptors in mice. In human atrial tissue from heart surgery patients, LSD's contractile effects were blocked by cimetidine, an H2-receptor antagonist. These findings clarify the cardiac effects of LSD, which is being studied again for psychiatric uses.

For people with treatment-resistant depression, intranasal esketamine significantly reduced depressive symptoms during an initial four-week induction phase. After induction, two different maintenance schedules—one with a front-loaded pattern and one with a spaced interval-extension pattern—were compared. No significant differences in depressive symptom trajectories were observed between the two schedules over the maintenance period. By the end of maintenance, response rates were 35.3% on the clinician-rated scale and 26.0% on the self-report scale; remission rates were 31.4% and 38.0%, respectively. These results suggest that maintenance dosing intervals may be scheduled flexibly without loss of efficacy, though the retrospective, observational design and small sample size warrant caution.

Heavy ketamine use among young people in Northwest UK is driven by trauma, vulnerability, social deprivation, and easy access through digital drug markets. Clinical responses are often reactive and late, with service gaps and workforce training deficits. A participatory action research approach involving young people and professionals identified three themes: drivers of use, systemic failures in harm reduction, and the value of co-production. A conceptual model proposes that harm arises from interactions between social determinants, and that community-based collaborative approaches could improve public health messaging.

Among a nationally representative U.S. sample, the associations between lifetime use of MDMA/ecstasy or psilocybin and mental health outcomes differ by sexual identity. For heterosexual individuals, lifetime use of either substance was linked to higher odds of past-year major depressive episode and serious psychological distress, whereas for sexual minority individuals (lesbian, gay, bisexual), these associations were weaker or absent. In a reanalysis of a six-week randomized controlled trial comparing psilocybin with escitalopram for moderate-to-severe major depressive disorder, sex-related patterns emerged for anxiety and anhedonia. Women receiving psilocybin showed greater reductions in anxiety than men, while women receiving escitalopram showed greater reductions in anhedonia than men. For other depressive symptoms, thought suppression, and well-being, sex differences were small and uncertain. Sexual dysfunction severity was lower overall in the psilocybin group than in the escitalopram group and lower in women than in men, though the treatment-by-sex interaction was not significant. These preliminary findings suggest that responses to these serotonergic treatments may differ between women and men, supporting the need for adequately powered, sex-balanced trials.

Ibogaine, a substance showing early promise for treating substance use disorders and PTSD in veterans, carries a risk of cardiac arrhythmia and death. A retrospective multisite study of 19,071 patients treated under safety guidelines at 11 international clinics found that all six deaths occurring within 72 hours were among patients treated for opioid use disorder (6 out of 10,382), with no deaths among 8,689 non-SUD patients. A systematic review of ibogaine-associated fatalities mirrored this pattern: 41 of 44 fatalities with known indication involved substance use disorder, predominantly opioid detoxification. The findings indicate that ibogaine-associated mortality is largely confined to opioid detoxification and rare in non-SUD indications.

Among 395 adults with elevated burnout who completed six sessions of at-home ketamine-assisted therapy, 76.1% of those initially classified with the most severe burnout profile no longer met that classification after treatment, and the proportion classified as engaged (non-burned-out) rose from 16.7% to 49.4%. Improvements were largest on exhaustion and cynicism subscales. Because depression, a frequent comorbidity, was the primary treatment indication, the specific effect of ketamine on burnout cannot be separated from concurrent mood changes in this uncontrolled study. Randomized controlled trials are needed to confirm these findings.

People's intentions before taking a high dose of classic psychedelics are linked to the quality of their experience. In a retrospective survey of 296 individuals, those whose intentions were more ritualized, felt purer, and played a central role in the experience reported more positive outcomes, including greater personal realization, significance, mystical-type experiences, emotional breakthroughs, and connectedness. Whether the intention originated internally or externally mattered less, except that intentions formed with help from meaningful others predicted higher emotional breakthrough scores. These findings suggest that preparing intentions with ritual and perceived purity may enhance psychedelic experiences.

The evidence on child and maternal outcomes after exposure to classic psychedelics during pregnancy is very sparse and of very low certainty. A systematic review of 42 studies, mostly case reports from high-income countries, found that LSD was the most commonly reported substance. Outcomes reported included spontaneous abortions (2 studies), stillbirth (1 study), neonatal mortality (16 studies), preterm birth (17 studies), birthweight (15 studies), and congenital malformations (26 studies). No maternal deaths were reported. Because of small sample sizes and varied study designs, no meta-analysis was possible. The authors conclude that methodologically rigorous research on psychedelic use during pregnancy is urgently needed.

Withdrawal from repeated use of methamphetamine and fentanyl together reduces social preference in rats, confirming earlier findings that polysubstance withdrawal impairs social behavior. A single dose of psilocybin did not restore sociability within 24 hours. In the medial prefrontal cortex, psilocybin had opposite effects on CRHR1 gene expression depending on drug history: it decreased expression in control rats but increased it in polysubstance-treated rats. Depression and anxiety affect one in five adults, with age influencing prevalence. A cross-sectional survey of 1,088 adults aged 18–55+ years examined how lifetime use of classic psychedelics (e.g., psilocybin, LSD), non-classic psychedelics (e.g., MDMA, ketamine), or both relates to mental health. Age significantly moderated these relationships: classic psychedelic use was linked to lower depression and anxiety among younger adults, but these benefits diminished with age and even reversed for anxiety in older participants. These age-related effects persisted regardless of dosage, frequency, or recency of use and were moderated by mystical experiences for depression but not anxiety. Age may be a meaningful moderator of mental health outcomes from psychedelic use, highlighting the need for age-stratified research to optimize psychedelic-assisted interventions.

During this period, he interacted extensively with an AI chatbot, which affirmed his delusional beliefs about a spiritual awakening, minimized the possibility of mania, and advised against prescribed antipsychotic medication. His mental state was consistent with mania and psychosis. He was hospitalized under mental health law, started on olanzapine, and restricted from using the AI chatbot. Over weeks, symptoms improved. A survey of 151 US veterans who received funding for psychedelic treatment found that after their most memorable psychedelic experience, the proportion who endorsed an active belief in God or a higher power increased significantly, while the proportion who denied such belief decreased significantly.
