Psilocybin-assisted therapy for depression: A systematic review and dose-response meta-analysis of human studies

A systematic review and dose-response meta-analysis of seven double-blind randomized placebo-controlled trials involving 489 adults with depression found that the optimal daily dose of psilocybin to reduce depression scores varies by population. The 95% effective dose (ED95) was 8.92 mg/70 kg for secondary depression, 24.68 mg/70 kg for primary depression, and 36.08 mg/70 kg when combining both subgroups. Dose-response associations were significant for all groups except a bell-shaped curve appeared for secondary depression. Higher doses were linked to increased side effects including physical discomfort, blood pressure increase, nausea, headache, and risk of prolonged psychosis. The analysis indicates that treatment-resistant depression requires higher doses than primary or secondary depression.

Half a Century of Research for Mind–Body Interventions: A Scientometric Analysis

A scientometric analysis of 16,310 documents from 1973 to 2023 reveals that mindfulness dominates mind–body intervention research, but each practice—Yoga, Qi-gong, Tai-chi, Vipassana, Zen, Loving-kindness, and Transcendental meditation—has distinct research clusters focused on different mental and physical health issues, such as substance abuse, schizophrenia, back pain, and dementia. The findings suggest that future research should recognize the unique contributions of each practice rather than treating them as a single category.

Treatment approaches and efficacy in psychedelic-induced psychosis: A systematic review.

Psychedelic-induced psychosis is rare, occurring in less than 1% of users in controlled trials, but evidence on its treatment is limited. A systematic review of 93 cases from 1955 to 2024 found that LSD (47.3%) and MDMA (38.7%) were the most common substances involved, with an average patient age of 23.7 years and 88% male. Psychosis lasted about 1.8 weeks on average. Second-generation antipsychotics had a response rate of 91.3%, significantly higher than first-generation antipsychotics at 27%. Electroconvulsive therapy also showed a 91% response rate. Follow-up revealed 34% of patients later developed schizophrenia spectrum disorders and 20.4% bipolar disorder, though limited follow-up data constrain these findings.

Treatment of depressive and negative symptoms in individuals with schizoaffective disorders using serotoninergic psychedelics: a case report.

Mindfulness-based interventions for patients with schizophrenia spectrum disorders: A systematic review of the literature.

A systematic review of 22 randomized controlled trials involving 1,500 patients with schizophrenia spectrum disorder (SSD) examined the structure of mindfulness-based interventions (MBIs). All programs varied in session components, duration, and instructor experience. MBSR-like programs focused on stress reactivity, while MBCT-like programs addressed primary psychosis symptoms and relapse prevention. Common mechanisms included attention training, emotion and stress regulation, decentering, self-compassion, and cognitive restructuring. The heterogeneity limits interpretation, but most recent trials had lower bias risk and more homogenous programs. Findings suggested potential benefits such as reduced negative symptoms, increased well-being, and decreased hospitalization rates. Authors recommend aligning on more congruent MBI programs for SSD and call for further research on optimal teaching approaches, mechanisms, and doses.
