Oral Ketamine for the Treatment of Depression: A randomized controlled trial and meta-analysis

medRxiv Preprint Server March 21, 2025 Leo R. Silberbauer, Benjamin Eggerstorfer, Paul Michenthaler et al. 1 citation preprint

Background Ketamine represents a significant advancement in antidepressant therapy, but the commonly used intravenous and intranasal application routes currently limit its availability beyond specialized centers. By contrast, oral ketamine treatment might constitute an alternative that is easy to administer with established safety and efficacy for patients with chronic and severe pain at home.

Associations between hypothalamic-pituitary-adrenal (HPA) axis hormone levels, major depression features and antidepressant effects of ketamine.

Journal of Affective Disorders March 15, 2025 Polymnia Georgiou, Cristan A Farmer, Gustavo C Medeiros et al. 24 citations

Subanesthetic doses of (R,S)-ketamine (ketamine) have demonstrated rapid and robust antidepressant effects in individuals with depression. However, individual variability in response to ketamine exists, and current biomarkers of ketamine treatment response are not entirely understood. Preclinical evidence suggests a link between hypothalamic-pituitary-adrenal (HPA) axis activation, a...

Trends and characteristics in ketamine use among US adults with and without depression, 2015-2022.

Journal of Affective Disorders March 15, 2025 Kevin H. Yang, Wayne Kepner, Charles M Cleland et al. 19 citations

Ketamine's potential for treating depression has drawn increased clinical interest in recent years. However, despite growing therapeutic use, recreational use among individuals with depression remain underexplored. We analyzed data from the 2015-2022 National Survey on Drug Use and Health focusing on adults in the US. Trends in past-year ketamine use, overall and by depression status, were...

Person-centred care during treatment with nasal esketamine - a qualitative study.

BMC Nursing March 14, 2025 Lina Klysing, Ingrid Larsson, Katrin Häggström Westberg 2 citations

Patients suffering from treatment-resistant depression may be treated with nasal esketamine. The treatment requires administration in a healthcare facility and monitoring by a nurse. Existing research has mainly focused on the clinical effect of nasal esketamine, while less is known about patients' experiences of treatment. A person-centred framework provides valuable insights into care and...

Effect of Esketamine on Depressive Symptoms in Adolescents With Major Depressive Disorder at Imminent Suicide Risk: A Randomized Psychoactive-Controlled Study.

Journal of the American Academy of Child and Adolescent Psychiatry March 7, 2025 Colette Kosik-Gonzalez, Dong-Jing Fu, Li Nancy Chen et al. 14 citations

To evaluate the efficacy, safety, and tolerability of esketamine nasal spray vs psychoactive placebo (oral midazolam) in rapidly reducing depressive symptoms in adolescents with major depressive disorder at imminent risk for suicide. This double-blind, double-dummy, phase 2b study randomized (1:1:1:2) 147 adolescents (12 to <18 years old) to esketamine (28, 56, or 84 mg) or midazolam twice...

Effect of esketamine on reducing postpartum pain and depression.

World journal of clinical cases March 6, 2025 Brandon Lucke-Wold, Armin Karamian 9 citations

In this editorial, we comment on a recent article by Chen et al, that addressed the effect of intraoperative injection of esketamine on postoperative analgesia and postoperative rehabilitation after cesarean section. Poor management of post-cesarean pain is associated with decreased maternal care for the baby, longer hospitalization, and higher risk of developing postpartum depression....

The Impact of Ketamine-Based Versus Non-Ketamine-Based ECT Anesthesia Regimens on the Severity of Patients’ Depression and Occurrence of Adverse Events: A Systematic Review with Meta-Analysis

Annals of Pharmacotherapy March 1, 2025 Dakota Sicignano, Rohan Kantesaria, Matthew Mastropietro et al.

Objective: To compare efficacy and safety outcomes for ketamine anesthesia + electroconvulsive therapy (ECT) versus nonketamine anesthesia + ECT in treatment-resistant depression (TRD) patients. Data Sources: PubMed and Embase were searched from the earliest date through November 27, 2023. Perspectives in treatment-resistant depression: esketamine and electroconvulsive therapy.

Wiener klinische Wochenschrift March 1, 2025 Pia Baldinger-Melich, Marie Spies, Ina Bozic et al. 5 citations

Modern electroconvulsive therapy (ECT) and the approval of nasal esketamine for clinical use have significantly improved the approach to treatment-resistant depression (TRD), which is defined as non-response to at least two different courses of antidepressants with verified adherence to treatment, adequate dosage, and duration of treatment. The goal of this literature review is to present the...

Clinical indicators of the suicide crisis and response to ketamine.

Journal of Affective Disorders March 1, 2025 Elizabeth D. Ballard, Lucinda Neely, Laura Waldman et al. 4 citations

This analysis sought to identify potential clinical targets for the suicide crisis. Characteristics of a useful clinical target include elevation at the time of suicide crisis and responsiveness to rapid-acting interventions. Suicidal ideation (SI), depression, and hopelessness were hypothesized to meet these criteria. Participants were 118 adults across the continuum of suicide risk, including...

Effect of continuous esketamine infusion on brain white matter microstructure in patients with major depression: A diffusion tensor imaging study.

Journal of Affective Disorders March 1, 2025 Xiang Liu, Zhipeng Wei, Lifeng Li et al. 12 citations

Esketamine has demonstrated acute antidepressant effects in patients with major depressive disorder (MDD). This study investigated whether these effects associate with reversible white matter fiber integrity recovery using diffusion imaging. Twenty patients with MDD and 20 healthy controls received 2-week esketamine treatment. Patients received 0.25 mg/kg intravenous esketamine. Emotional and...
