Sex differences in placebo and antidepressant response to intranasal esketamine for treatment-resistant depression

Molecular Psychiatry February 18, 2026 Marie Huc, Sara Siddiqi, Mysa Myers et al. 1 citation

Esketamine has emerged as a fast-acting antidepressant option for individuals with treatment-resistant depression (TRD). Yet, little is known about how sex-assigned-at-birth shapes symptom-specific responses to these interventions, a critical gap in the move toward precision psychiatry. To address this gap, we conducted a pooled analysis of five randomized, double-blind, placebo-controlled...

LSD microdosing for major depressive disorder: Mood and pharmacokinetic outcomes from a Phase 2a trial

Progress in Neuro-Psychopharmacology and Biological Psychiatry February 18, 2026 Dimitri Henriques Daldegan-Bueno, C Donegan, Rachael Sumner et al. 1 citation

Results suggest short-term improvements in mood following microdosed LSD in people with depression, warranting confirmation in controlled trials. It provides the pharmacokinetic parameters of 8 μg of LSD in a sample of people with depression and indicates no tolerance or sensitisation to repeated microdoses of LSD, despite incremental dose titration.

The Antioxidant Activity of Ketamine: Threshold-Dependent Mechanism in Treatment-Resistant Depression?

Research Square February 17, 2026 Zofia Winczewska, Magdalena Górska-ponikowska, Wiesław Jerzy Cubała preprint

Abstract Aim of study: The antidepressant effect of ketamine may be at least partly due to its antioxidant activity. The aim of the study was to assess whether ketamine develops a protective effect on HT22 neuronal cells under conditions of oxidative stress (OS) induced by hydrogen peroxide (H 2 O 2 ). Material and methods The experiment was performed on an immortalized mouse hippocampal HT22...

A short-acting psychedelic intervention for major depressive disorder: a phase IIa randomized placebo-controlled trial.

Nature Medicine February 16, 2026 David Erritzøe, Tommaso Barba, Tiffanie Benway et al. 4 citations

Major depressive disorder (MDD) is a leading cause of disability worldwide, yet many patients have inadequate responses to current treatments. Dimethyltryptamine (DMT), a serotonergic psychedelic with rapid onset and short duration, shows promise as a potential antidepressant (AD), although clinical evidence in MDD remains limited. We conducted a phase IIa, double-blind, placebo-controlled,...

Microdosing psilocybin for major depressive disorder: study protocol for a phase II double-blind placebo-controlled randomised partial crossover trial

BJPsych Open February 16, 2026 Zeina Beidas, Anya Ragnhildstveit, Adam Blackman et al. 1 citation

BACKGROUND: Major depressive disorder (MDD) is the leading cause of disability worldwide, affecting roughly 322 million people. Recently, doses of psilocybin have shown promise in treating mood disorders, sparking interest in other dosing practices. According to anecdotal reports and observational studies, microdosing psilocybin yields benefits to mental health; however, rigorously controlled...

ADVERSE EFFECTS OF ESKETAMINE IN TREATMENT RESISTANT DEPRESSION: A COMPREHENSIVE LITERATURE REVIEW (2020-2025)

International Journal of Innovative Technologies in Social Science February 16, 2026 Aleksandra Lejman, Rafał Bednarczyk, Natalia Bednarczyk et al.

Background: Esketamine nasal spray represents the first FDA approved treatment with a novel mechanism of action for treatment resistant depression (TRD) in decades. While its efficacy has been well established, a thorough understanding of the adverse effect profile remains essential for informed clinical decision making and optimal patient safety. We therefore undertook this systematic review...

Does number of dosing sessions and type of adjunctive psychotherapy affect outcomes in psychedelic-assisted-psychotherapy for depressive disorders and anxiety disorders? A systematic review and meta-analysis.

Journal of Affective Disorders February 15, 2026 Tomas Lindegaard, Anton Käll

Although there is increasing evidence of the efficacy of psychedelic-assisted psychotherapy (PAP) for depressive and anxiety disorders, less is known regarding how different methodological aspects affect outcomes. This study investigated the moderating impact of one or multiple dosing sessions and type of adjunctive psychotherapy on the efficacy of PAP in depression and anxiety populations....

Ketamine ameliorates postpartum depression-like behaviors in rats exposed to sevoflurane during pregnancy through the AMPK/SIRT1/NLRP3 pathway.

Journal of Affective Disorders February 14, 2026 H. Xue, Xu Yang, Shihui Kuai et al.

General anesthetics can exert significant adverse effects on the central nervous system. This study aimed to investigate whether repeated exposure to sevoflurane induces depression-like behaviors in postpartum rats. Pregnant rats were exposed to 3% sevoflurane for 2 h on gestational days 13-15. Emotional behaviors were assessed on postpartum days 1, 7, 14, and 21. Hippocampal protein levels...

Network localization of brain functional effects of ketamine treatment for major depression.

European Psychiatry February 13, 2026 Haining Ma, Huaigui Liu, Wenwei Zhang et al.

Considerable effort has been devoted to investigate the neuroimaging correlates and predictors of antidepressant response to ketamine, yet inconsistency in the location and nature of the regional brain effects makes it difficult to unify this research. Despite the revolutionary notion that psychiatric therapeutics show network-level brain representations, investigations into network...

Time-Dependent Effects of Rapid-Acting Antidepressants in iPSC-Derived Neurons from Treatment-Resistant Depression and Healthy Volunteers.

Research Square February 12, 2026 Jenessa N Johnston, Greg Jones, Shiyong Peng et al. preprint

Rapid-acting antidepressants like ketamine and serotonergic psychedelics show promise for treatment-resistant depression (TRD), but the molecular mechanisms that contribute to their therapeutic effects remain unclear. Induced pluripotent stem cells (iPSCs) offer a platform to model human cortical neurons and investigate drug effects in a human-relevant system. Here, iPSCs from individuals with...
