Short term changes in the proteome of human cerebral organoids induced by 5-MeO-DMT.

5-MeO-DMT, a serotonin-like molecule found in traditional Amerindian medicine, alters the protein landscape of human cerebral organoids. Out of 6,728 identified proteins, 934 were differentially expressed after treatment. Computational analysis confirmed previously reported anti-inflammatory effects and revealed modulation of proteins involved in long-term potentiation, dendritic spine formation, cellular protrusion, microtubule dynamics, and cytoskeletal reorganization. These findings provide initial molecular insights into how this compound may affect human brain metabolism.

Religions have long recognized lucid dreaming (LD) as an important conscious state, predating modern scientific study by millennia. Hindu texts over 2,000 years old divide consciousness into waking, dreaming (including LD), and deep sleep. Tibetan Buddhists practice Dream Yoga to recognize dreams, overcome fears, and control dream content. Islam regards LD as a valuable mental state for mystical experiences. Christian theologian Augustine described LD as a preview of the afterlife. Spiritism in the nineteenth century linked LD to out-of-body experiences. Abrahamic religions view dreams as communication with God, while Indian traditions cultivate self-awareness through LD induction techniques. These historical religious insights can inform current scientific research on LD phenomenology.

An international Delphi consensus for reporting of setting in psychedelic clinical trials.

A Delphi consensus study with 89 experts from 17 countries identified 30 extra-pharmacological variables that are important or very important for reporting the setting in psychedelic clinical trials. These variables, forming the ReSPCT guidelines, cover physical environment, dosing session procedure, therapeutic framework and protocol, and subjective experiences. The findings reveal significant ambiguities in current conceptualizations of set and setting. The guidelines provide a new standard for designing and documenting contextual factors in psychedelic research.

Information parity on cortical functional brain networks increases under psychedelic influences

The physical basis of consciousness remains an open question. Using complex network theory, the study compared resting-state functional brain networks of individuals before and after ingesting the psychedelic brew Ayahuasca. The researchers calculated pairwise information parity to quantify statistical symmetries between brain region connectivity across the entire network. They detected an increase in average information parity in brain networks under psychedelic influence. Notably, information parity between regions of the limbic system and frontal cortex was consistently higher for all individuals while under the influence.

Characterizing complex networks using Entropy-degree diagrams: unveiling changes in functional brain connectivity induced by Ayahuasca

A new network metric called geodesic entropy quantifies the Shannon entropy of the distance distribution from each node to all others, capturing the structural role of individual nodes in a network. Applied to functional brain networks of humans in two states of consciousness—ordinary waking and after ingestion of Ayahuasca—the metric distinguished the states. Brain networks under Ayahuasca showed, on average, larger geodesic entropy than those in the ordinary waking state. The authors suggest geodesic entropy is a useful tool for analyzing complex networks and may improve understanding of emergent behaviors such as self-organized criticality.

Psychedelic Ayahuasca increases brain network complexity, supporting ancient wisdom about "mind expansion" with modern neuroscience. Brain scans revealed that this Amazonian brew creates more diverse neural connections while strengthening local brain networks. The changes in brain organization showed higher Shannon entropy, indicating more dynamic and flexible thought patterns during the psychedelic experience.
