Abstract
Endogenous N,N-dimethyltryptamine (DMT) has been detected in mammals, but its physiological functions and the distribution of functionally relevant synthesis remain unresolved. This paper proposes prostatic tissue as a candidate peripheral site of synthesis. Oncology studies document indolethylamine N-methyltransferase (INMT) expression in human prostate samples, including adjacent non-cancerous tissue, without establishing tryptamine synthesis there. It is hypothesised that prostatic tissue produces N,N-DMT and/or 5-methoxy-DMT (5-MeO-DMT), that production increases during specified conditions of pelvic autonomic activation, and that local tryptamine action modifies prostate-associated afferent signalling to the central nervous system. This model does not require transport of the molecule to the brain. The two analytes require separate biosynthetic and pharmacological assessment. The experimental programme separates tissue detection, demonstration of local biosynthesis, statedependence, and causal neural effects. Targeted LC-MS/MS is the first screening step; positive detection alone would not establish synthesis. Negative results would constrain or disconfirm specified claims only under validated sampling and assay conditions. The proposal is a testable biochemical and neurophysiological hypothesis, not an established explanation of altered experience.
