Zusammenfassung Psilocybin (4‐Phosphoryloxy‐ N , N ‐dimethyltryptamin, 1 ) ist der hauptsächliche Indolethylamin‐Naturstoff der psychotropen sogenannten Zauberpilze. Die Mehrheit der 1 ‐produzierenden Arten gehört zu der namensgebenden Gattung Psilocybe , für die die biosynthetischen Ereignisse, ausgehend von l ‐Tryptophan ( 2 ), sowie die beteiligten Enzyme gründlich charakterisiert wurden. Einige Inocybe ‐Arten (Risspilze), darunter Inocybe corydalina , produzieren ebenfalls 1 . In Produktbildungsassays charakterisierten wir vier rekombinant hergestellte Biosyntheseenzyme dieser Art in vitro: IpsD, eine Pyridoxal‐5′‐phosphat‐abhängige l ‐Tryptophandecarboxylase, die Kinase IpsK, sowie zwei fast identische Methyltransferasen, IpsM1 und IpsM2. Das fünfte Enzym, die unlösliche Monooxygenase IpsH, wurde in silico untersucht. Erstaunlicherweise findet keine der Reaktionen des 1 ‐Biosynthesewegs in Psilocybe ‐Arten in I. corydalina statt. Im Gegensatz zur Situation in Psilocybe ist der Inocybe ‐Biosyntheseweg verzweigt und führt zu Baeocystin (4‐Phosphoryloxy‐ N ‐methyltryptamin, 3 ) als zweitem Endprodukt. Unsere Ergebnisse zeigen, dass Pilze nur entfernt oder nicht verwandte Enzyme verwendeten, um die metabolische Fähigkeit der 1 ‐Biosynthese zweimal unabhängig zu evolvieren.
