Co-administration of cannabidiol and ketamine induces antidepressant-like effects devoid of hyperlocomotor side-effects.

In male Swiss mice, cannabidiol (CBD) produced dose-dependent antidepressant-like effects without causing hyperlocomotion, whereas S-ketamine produced antidepressant effects alongside hyperlocomotion at higher doses. Blocking AMPA receptors prevented the antidepressant effects of both compounds. Pretreatment with CBD (10 mg/kg) reduced ketamine-induced hyperlocomotion while preserving its antidepressant effect. These findings suggest that combining CBD with ketamine may achieve antidepressant benefits with fewer psychostimulant side effects.
